Furthermore, confirmation during specific stages of gestation and at dry-off holds significant economic importance.
Traditionally, pregnancy in cattle within regions like Nepal has been determined through manual rectal palpation or by transrectal ultrasound (TU). While these methods are widely practiced, they also present distinct operational constraints, physical demands, and potential stress factors for the animals involved.
Overview of Bovine Pregnancy Rapid Test Kit
To address the need for more efficient and accessible diagnostic tools, Novala Biotech has developed a Bovine Pregnancy Rapid Test Kit. This product is a lateral flow immunoassay designed for the qualitative detection of Pregnancy-associated glycoproteins (PAG) in bovine whole blood, serum, or plasma.
Pregnancy-associated glycoproteins (PAG) serve as critical detection markers for early pregnancy testing in bovines. These glycoproteins begin to appear in the blood around 25 days after mating and can be reliably detected after 28 days using the test cassette. The test provides users with a method for accurate detectionas early as 28 dayspost-breeding and throughout gestation, and it can be utilized 70 days or more after calving.
Advantages Over Manual Rectal Palpation
Transitioning from traditional manual methods to rapid immunological test kits offers several critical advantages for livestock health and farm workflow:

Key Information Regarding Usage of the Product
Proper storage, specimen handling, and testing execution are critical to obtaining accurate diagnostic results.
Storage and Stability
- Temperature Range: Keep the test kit between 4–30°C in a dry, cool place and avoid freezing.
- **Shelf Life:**The kit remains stable within the expiry date printed on the product label and outer packaging; do not use past this date.
- **Packaging:**Packages should remain sealed until required for use.
Specimen Collection and Preparation
- Specimen Types: Whole blood, serum, or plasma. Whole blood specimens should contain EDTA, citrate, or heparin, collected by vein puncture following standard procedures. Whole Blood can also be used immediately after drawing if reagents are unavailable.
- Hemolysis Prevention: Separate serum or plasma from blood as soon as possible to avoid hemolysis; use only clear, non-hemolyzed specimens. (Note: The use of plasma or serum specimens is highly recommended).
- Storage Conditions: Store specimens at 2–8°C for up to 7 days if not testing immediately. For longer storage, freeze specimens at -20°C. Do not freeze whole blood specimens.
- Sample Treatment: Avoid multiple freeze-thaw cycles. Bring frozen specimens to room temperature slowly and mix gently prior to testing. Clarify specimens containing visible particulate matter by centrifugation.
Test Procedure
Allow the test device and specimens to equilibrate to 15–30°C prior to testing, place on a clean level surface, and follow the procedure below:

Result Interpretation and Quality Control
- Two Pink Lines (Positive): Indicates the presence of PAG, confirming pregnancy.
- One Pink Line at C (Negative): Indicates PAG is absent or below detectable levels.
- No Pink Line (Invalid): Indicates improper procedure or membrane failure.
- Quality Control: A colored line appearing in the control region (C) acts as an internal procedural control, confirming sufficient specimen volume, adequate membrane wicking, and correct technique.
